To clarify the influence of soft laser irradiation on gene expression of epidermal growth factorreceptor(EGFR)和extra cellular matrix in epithelial cells were examined. Normal human epithelialkeratinocytes(NHEK) were cultured in a 24 well microplate(5xlo - D13 - D1cell/cm - D12 - D1),和laser device was used Trnpl D(GaAlAs diode, 905nm wave length, max out put 10w, average 2.4mW,energy density 1.25J/cm (150sec.))(Yoshida,Japan). NHEK were irradiated for 20 sec. From the bottom of plate after incubated for 48hrs. EGFR表pression was investigated immunohistochemically and in situ hybridization. Fibronectin and lamininELISA. Also integrin α2β1keratin and actin filament of NHEK were examined.After laser irradiation,proliferation of NHEK was increasedactivity and gene expression of EGFR was accelerated. Fibronectin in the medium showed no重要差异compered with control虽然laminin in the medium was decreased after laser irradiation. To clarify the influence of soft laser irradiation on gene expression of epidermal growth factor receptor(EGFR) and extra cellular matrix in epithelial cells were examined. Normal human epithelial keratinocytes(NHEK) were cultured in a 24 well microplate(5 X lOィイD13ィエD1cell/cmィイD12ィエD1), and laser device was used Trnpl D(GaAlAs diode, 905nm wave length, max out put 10W, average 2.4mW, energy density 1.25J/cmィイD12ィエD1(150sec.))(Yoshida, Japan). NHEK were irradiated for 20 sec. From the bottom of plate after incubated for 48hrs. EGFR expression was investigated immunohistochemically and in situ hybridization. Fibronectin and laminin were mesured by ELISA. Also integrin α2β1, keratin and actin filament of NHEK were examined.After laser irradiation, proliferation of NHEK was increased, activity and gene expression of EGFR was accelerated. Fibronectin in the medium showed no significant difference compered with control, although laminin in the medium was decreased after laser irradiation. On the other hand, integrin expression was activated, also keratin and actin filaments of NHEK were developed upon laser irradiation.These results suggest that laser irradiation activate EGFR of NHEK, so cell proliferation may accelerate, and activation of integrin and extra cellular matrix expression by laser irradiation may promote both cell adhesion and cell differentiation.
