Using op/op mice without the macrophage-colony stimulating factor (M-CSF) activity and litter normal mice with the M-CSF activity, comparison examination on the bone resorption of HDP particles induced was carried out.Burr hole was produced in the parietal bone of these 2 groups, and the high density polyethylene particles (HDP, 10 μm diameter or less) were transplanted. The graft place was exposed after the selected time period, and the observation by H.E staining, identification of osteoclast by tartaric acid resistance acidity phosphatase (TRAP) staining and macrophage by F4/80 and Mac2 antimouse macrophage antibody and transmission electron microscopic observation were carried out. In the H.E staining, the mononuclear cells in which normal mice, op/op mice also took in HDP particles particles were recognized in great numbers. In the normal mice, anyway showed positive, when F4/80, Mac2 antibody staining and TRAP staining were observed (however, the TRAP positive cell are very few). In the op/op mice, though the Mac2 antibody staining was the positive, the positive cells could not be together recognized F4/80 antibody staining and TRAP staining. In the electron microscopic observation, the cells which took in HDP particles showed clearly morplological features of the fibroblast. And the uptakes of HDP particles by macrophage was also able to be observed in the normal mice.From present examination, it was proven that not only macrophage but also fibroblast also actively take in HDP particles, and it was indicated that fibroblast played an important role for the bone resorption. And TRAP positive, mononuclear cell which took in HDP was observed, there is no report which osteoclast or pre-osteoclast have phagocytosis, so that is a new knowledge, when the role of osteoclast in the bone resorption is considered.
采用无巨噬细胞集落刺激因子(M-CSF)活性的op/op小鼠和有M-CSF活性的正常小鼠,对HDP颗粒诱导的骨吸收进行比较检查,在这2组小鼠的顶骨中产生毛刺孔,并移植高密度聚乙烯颗粒(HDP,直径10 μm或更小)。在选定的时间段后暴露移植部位,进行H.E染色观察,抗酒石酸酸性磷酸酶(TRAP)染色鉴定破骨细胞,F4/80和Mac 2抗小鼠巨噬细胞抗体鉴定巨噬细胞,透射电镜观察。在H.E染色中,正常小鼠、OP/OP小鼠的单个核细胞也大量摄取HDP颗粒。正常小鼠F4/80、Mac 2抗体染色及TRAP染色均呈阳性(但TRAP阳性细胞极少)。在op/op小鼠中,虽然Mac 2抗体染色呈阳性,但阳性细胞不能被F4/80抗体染色和TRAP染色共同识别。电镜观察,摄取HDP颗粒的细胞具有明显的成纤维细胞形态学特征。在正常小鼠中也能观察到巨噬细胞对HDP颗粒的摄取,证明不仅巨噬细胞而且成纤维细胞也能主动摄取HDP颗粒,表明成纤维细胞在骨吸收中起重要作用。而TRAP阳性的单个核细胞可吞噬HDP,破骨细胞或破骨前体细胞具有吞噬作用尚未见报道,因此,当考虑破骨细胞在骨吸收中的作用时,这是一个新的认识。
