Five-week-old syngeneic BL/6 mouse served as donors for Lac-Z transgenic mouse. Bone marrow transplantation was performed.After whole body irradiation with 900cOy. Fibrotic liverwas induced by administration of OCL4. Hepatocyle growth factor(HGF) gene transfer was performed and examined if bone marrow cell would be delivered to liver or not in regeneration of liver.We got the following finds. Hepatocytes or oval cells did not derive from hematopoietic stem cell in hepatic regeneration in a little damaged liver. After hematopoietic stem cell was delivered to the endothelial cell, the liver was reconstructed. In this experiment, the co-lateral vein developed but hematopoietic cell was not found. We could not show the mechanism of the development of co-lateral vein in portal hypertension.
取出生5周的同基因BL/6小鼠作为Lac-Z转基因小鼠的供体。经900cOy全身照射后进行骨髓移植。OCl4诱导大鼠肝纤维化模型。进行肝细胞生长因子(HGF)基因转移,检测骨髓细胞是否能进入肝脏再生。在轻度肝损伤的肝再生过程中,肝细胞或卵圆细胞不是来源于造血干细胞。当造血干细胞被输送到内皮细胞后,肝脏被重建。在本实验中,共侧静脉发达,但未发现造血细胞。尚不能揭示门静脉高压症同侧静脉形成的机制。
