项目摘要

1.口腔乾燥症モデル動物の確立10週齢のウィスター系雄性ラット40匹(10匹×4群)を用いた.腹腔内麻酔下において唾液腺の摘出を行った.1群には,耳下腺導管の結紮を行い.また,1群には,耳下腺導管結紮および,顎下腺,舌下腺の摘出を行う.その他の1群には偽手術を行い,残りの1群は無処置のまま経過させた.手術5週後から,1週間ごとに唾液量(ろ紙を用いて測定),水分摂取量,食物摂取量,行動の変化を観察した.手術10週後にすべての動物を屠殺し,口蓋組織,歯周組織舌を採取した.採取した組織はヘマトキシリン-エオジン染色を施した.組織観察の結果と唾液分泌量他の所見を考慮すると,モデル動物としては,耳下腺導管結紮,顎下腺・舌下腺摘出を行った動物が最も適しており,術式としても問題なく実験に供することができると判断された.2.実験的口腔乾燥症下における義歯床下粘膜のダメージと治癒過程の観察10週齢のウィスター系雄性ラット125匹(25匹×5群)を用いた.耳下腺結紮・顎下腺・舌下腺摘出により口腔乾燥症ラットを75匹作製した.それらのうち2群の動物には,咀嚼圧を加える実験用義歯を装着した(13μm,50μm沈下群).残りの1群は実験用義歯を装着せずに経過させた.また,1群25匹の動物には偽手術を施し,残りの1群25匹は無処置のまま経過させた.手術5週後に実験用義歯を装着し,義歯装着1週後から1週間隔で5週後までを観察期間とした.各観察期間の終了時に,各実験群の各5匹を環流固定して,義歯床下の口蓋組織を採取した.採取した組織は固定,脱灰ののち,パラフィン包埋し,ヘマトキシリン-エオジン染色およびAgNOR染色を施した.組織観察ならびに評価・検討は来年度の予定となっている.
1. The animals of xerostomia were treated with 40 male animals (10 dogs x 4 groups).
