すでに作製済みのEA2の変異をもつES細胞をマイクロインジェクション法によりマウス初期胚に導入し、偽妊娠雌マウスの子宮に移植することでキメラマウスを得た。しかしながら、同様の方法を用いた他のノックインマウスで、変異を持つタンパクおよびRNAの発現が低いことがwestern blot法およびNorthern blot法により確認された。この原因として考えられる理由の1つに、ターゲティングベクターに組み込んでいたポジティブセレクション用のハイグロマイシン耐性遺伝子が影響している疑いが持たれている。そこで今回作製したEA2キメラマウスにおいても同様に、変異タンパクの発現が低い可能性が十分考えられることから、はじめからこの遺伝子を取り除くことにした。今回用いたターゲティングベクター上のハイグロマイシン耐性遺伝子はFRTサイトで挟んでおいたことから、FLPeのトランスジェニックマウスとこのハイグロマイシン耐性遺伝子を持つEA2キメラマウスを掛け合わせ、FLPe/FRT systemによりハイグロマイシン耐性遺伝子を取り除いた。この掛け合わせによって生まれたマウスのうち、EA2キメラ、ハイグロマイシン耐性遺伝子ネガティブ、FLPeポジティブのマウスをPCR法によってセレクトし、次にこのマウスをB6マウスと掛け合わせ、今度はEA2キメラ、ハイグロマイシン耐性遺伝子ネガティブ、FLPeネガティブのマウスを得ることができた。現在、EA2キメラでハイグロマイシン耐性遺伝子を持たないこのマウス同士を掛け合わせているところである。
