オキシコドンの脊髄クモ膜下腔投与は、投与量に依存して鎮痛効果を起こすが、脳幹毛様体への投与では鎮痛作用を観察しなかった。この知見をふまえて、PC12培養細胞において、オキシコドン単独やオキシコドンにNSAIDsを併用して、MAPK(mitogen-activated protein kinase)のERK活性を観察している。これと平行して、硬膜外腔に投与したモルヒネとオキシコドンで術後鎮痛と副作用を、下腹部手術を受ける患者に対し比較検討した。モルヒネと同量のオキシコドン投与では、術後鎮痛がモルヒネに及ばなかったが、悪心、嘔吐、掻痒感等の副作用の頻度は少なかった。また、モルヒネの2倍量のオキシコドン投与では、鎮痛効果はモルヒネと同等で、副作用は少ない結果であった。患者の満足度は、差はなかった。また、モルヒネと同量投与のオキシコドン単独ではモルヒネの鎮痛効果に及ばなかったが、オキシコドンにNSAIDsであるフルルビプロフェンを併用すると、鎮痛効果はモルヒネと同等になり、副作用はモルヒネ郡と比較して少ない結果であった。これらの結果は、2003年10月11-15日にサンフランシスコで行われた、アメリカ麻酔学会で発表した。
