これまで我々は、臍帯上皮には表皮に類似した構造をもつ領域が存在し、その領域においては表皮同様の分化関連蛋白の発現を認め、さらに臍帯から上皮細胞を分離培養し、コラーゲンゲル上で三次元培養を行うことにより、表皮角化細胞同様に重層化させることが可能であることを報告した。また、コラーゲンゲル上で単層培養した臍帯上皮細胞をヌードマウスの皮膚欠損部に移植したところ、角化細胞同様に10層前後に重層化し、また、各種ケラチン、角化の最終分化マーカー、基底膜構成蛋白に対する抗体を用いた免疫染色においても、表皮角化細胞とほぼ同様の染色パターンを示した。興味深いことに、移植直後の臍帯上皮細胞においては、表皮分化型ケラチンK1/10に加えて単純上皮型ケラチンK8/18及び粘膜型ケラチンK4/13を同時に発現しており、これらの発現は移植2週間後には減少していることが確認された。以上の結果から、臍帯上皮細胞は生体に移植することにより、in vitroと比較して、より表皮に近い分化を誘導することが可能であることが示唆された。以上のin vitro及びin vivoにおける臍帯上皮細胞の性状についてJournal of Dermatological Scienceに報告した。これらの結果は、臍帯上皮細胞を用いて表皮シートを作製し、臨床応用することが可能であることを示すものであり、現在、さらに移植後の長期的な観察を行っている。

This paper reports the existence and development of differentiation related proteins in the epidermis of human umbilical epithelium, the isolation and culture of epithelial cells in human umbilical epithelium, and the stratification of epidermal keratinocytes. In addition, the cells were cultured in layers, and the keratinocytes were transplanted into the damaged part of the skin. The keratinocytes were stratified before and after 10 layers. The keratinocytes were stained with antibodies against basement membrane proteins. In addition, epidermal differentiation type K1/10 and pure epithelial type K8/18 and mucosal type K4/13 were found simultaneously after transplantation. The above results show that the epithelial cells of umbilical cord can be transplanted in vivo. The above is reported in the Journal of Dermatological Science. This results in the use of epithelial cells in the control, clinical use, and long-term observation after transplantation.
