各皮膚悪性腫瘍細胞株に対するアンギオテンシンIIの影響を見た。前年度の実験で、AT1レセプターをもっていることを確認した皮膚腫瘍細胞株について検討した。アラマーブルーやBrdUの両評価法において、あらゆる濃度のアンギオテンシンIIを加えたときにも細胞の増殖能に有意な差は認めなかった。次に、一定濃度のアンギオテンシンII存在下におけるAT1阻害剤(10-5〜10-9mg/ml)、TA2阻害剤(10-4〜10-8mg/ml)の影響をみたが、これもどの濃度のときも細胞の増殖に有意な違いはみられなかった。使用した各阻害剤については、他の研究により最大効果を有することが確かめられた値を基に濃度を決定したが、期待した結果を得ることはできなかった。今後はさらに、作用時間や、阻害剤を変えて実験を進めたいと考えている。尋常性乾癬や円形脱毛症については、疾患の標本が入手できずに、実験を進めることはできなかった。しかし、アンギオテンシンIIが細胞増殖に関与していることは明らかであり、そのレセプターによってその作用が、細胞増殖であったり、アポトーシス誘導であったりすることも分かってきているので、常に細胞が増殖し続けている表皮や毛包において重要な役割を果たしていることは充分に考えられる。毛包に関しても、アポトーシスは毛周期にとって必須で、脱毛症の病因にアンギオテンシンIIが深く関与している可能性もある。今後も検討を続けていく予定である。
