Fish roe such as salmon, walleye pollack, and herring roes are popularly consumed in Japan and much cases of fish-roe-allergy, particularly in childhood, have been reported last decade. However; there is little information on fish roe allergens.The objective of this study is to identify fish roe allergens and to investigate the IgE cross-reactivity among various kinds of fish roes.The results were as follows: (1) Eleven kinds of fish roes (Churn salmon: Oncorhynchus keta, Sakhalin taimen: Hucho perryi, rainbow trout: Oncorlhynchus mykiss Japanese char: Salvelinus leucomaenis, dolly vuuden: Salvelinus malma, walleye pollack: Theragra chalcogramma, dusky sole Lepidopsetta mochigare, Slime flounder:Microstomus achne, atka mackerel: Plecurogrammus azonus, sishamo smelt: Spirinchus lanceolatus capelin: Mallotus villosus) contained yolk protein that reacted with anti chuun-salmon-β'-component rabbit IgG (anti-β) and they also reacted with specific IgE in chum salmon roe-allergic patients' se … More ra.(2) The major allergens in salmonid fish roes were β'-component and its related fragments.(3) There were IgE cross-reactivity among salmonid roe β'-components, and the IgE cross--reactivity was also confirmed between churn salmon and walleye pollack roes.(4) Amino acid sequence of churn salmon β'-component (Mw. 16,000) was examined and the sequences of 91 residues (about 60% of the total sequence) were determined by peptide mapping using the complete sequence of rainbow trout viltellogenin.(5) cDNA encoding β'-component was obtained using mRNAs extracted firm chum salmon liver and the encoded amino acid sequence was coincided with the sequence obtained by the direct peptide mapping.(6) β'-component like protein (Mw 17,000) was purified firm walleye pollack roe, and about 70% of the total amino acid sequence was determined using an inside arrangement of Atlantic cod vitellogenin as a template.These findings would contribute understanding fish roe allergy and improvement of QOL of allergic patients.
