The aim of this study is clarifying differential gene expression profiles and action mechanisms between genotoxic and non-genotoxic carcinogens in the target organ. The following results were obtained.1)Differential gene expression profiles of 2 N-nitroso carcinogenic compounds, diethylnitrosamine and dipropylnitrosamine, and a non-genotoxic carcinogenic compound, phenobarbital, in mouse liver at 4 h and 28 days after administration determined with DNA microarray and quantitative real-time PCR.2) Dose-dependent alteration in the gene expression in mouse liver induced by diethylnitrosamine and ethylnitrosourea at 4 h and 28 days after administration determined by quantitative real-time PCR.3) Time-dependent gene expression profiles at 4, 16, 20, 24, and 48 h after administration of a genotoxic carcinogen, chrysene, in mouse liver determined with quantitative real-time PCR.4) Differential gene expression profiles induced by 4 genotoxic carcinogens [quinoline, N-nitroso morpholine, 4-dimethyl (amino) azobenzene and 2, 4-diaminotoluene], and 2 non-genotoxic carcinogens (clofibrate and dichlorobenzene) in mouse liver at 4 and 48 h after administration determined with quantitative real-time PCR examined as training set.5) Differential gene expression profiles induced by 4 genotoxic carcinogens [3, 4-(methylnitrosamino)-a-(3-pyridyl)-1-butanone, 2-actylamonofluorene, urethane, diisopropanolnitrosamine] and 3 non-genotoxic carcinogens [di(2-ethylhexyl)phthalate, dichloro-diphenyl-trichloroethane and furan] in mouse liver at 4 and 48 h after administration determined with quantitative real-time PCR examined as validation set.
