项目摘要

ブタの背脂肪厚は枝肉の最も重要な評価項目の一つである。本課題では、アジア系梅山豚の背脂肪が西洋系品種に比べて厚い理由は脂肪細胞の元となる脂肪前駆細胞の増殖性に差異があるとの仮説を証明することを目的としている。当研究室で樹立した西洋系交雑豚由来の皮下脂肪前駆細胞(PSPA)と梅山豚由来の皮下脂肪前駆細胞(MSPA)のconfluent後10日目の増殖関連遺伝子の発現をアレイ解析により比較した結果、MSPAで2倍以上の高発現を示す9つの遺伝子が検出された。しかし、CDK6、CKS2、CCNB2、CCNFについては実際のアレイ上のスポットが両細胞区で視認できないほどのレベルであったため、残る5つの遺伝子についてリアルタイムRT-PCRにより再現性を調べた。BCL2は有意差が見られず、BRCA1はアレイの結果と逆転したが、CCNE2、CCNA1、CDC2の発現量はMSPAで有意に高いことを確認した。更に5つの遺伝子について、胎齢85日、生後1週、1ケ月、3ケ月及び5ケ月齢のランドレースと梅山豚から背脂肪を経時的に採取し、RNAを抽出、次いで合成したcDNAを用いて、リアルタイムRT-PCRによる発現量の比較を行った。ランドレース、梅山豚の成長に伴い、皮下脂肪組織における上記遺伝子の発現量はBCL2を除いて徐々に低減した。両品種共通の現象として胎児期から幼少期にかけて皮下脂肪における前駆細胞等の細胞増殖能が低下することが判明したが、個体差が大きいために品種間の違いを生体レベルで証明するには至らなかった。しかし、PSPAに比べてMSPAの方がG1期、S期及びM期に働く遺伝子の発現量が高いことから、西洋系品種よりも梅山豚由来の前駆細胞の方が細胞分裂を活発に行っていることが細胞レベルでは明らかになった。

The most important thing is to evaluate the project. The purpose of this study is to demonstrate the differences in the reproductive characteristics of adipocytes between Chinese and Western breeds of Meishan dolphin. Ten days after the laboratory established the confluent relationship between subcutaneous fat progenitor cells (PSPA) derived from the Western cross-breed dolphin and subcutaneous fat progenitor cells (MSPA) derived from the Meishan dolphin, we conducted a comprehensive analysis of the discovery of growth-related genes. The results showed that MSPA was more than twice as high as MSPA. Nine genes were detected. CDK6, CKS2, CCNB2, CCNF, CDK6, CKS2, CCNB2, CCNF3, CCNF4, CCNF5, CCNF6, CCNF5, CCNF6, CCNF7, CCNF8, CCNF9, CCNF9, CC BCL2 is intentionally high, BRCA1 is intentionally low, CCNE2, CCNA1, CDC2 is intentionally high, and MSPA is intentionally low. In addition, we compared the amount of DNA extracted, RNA extracted, cDNA synthesized and detected by RT-PCR at 5 days of gestation, 85 days of gestation, 1 week, 1 month, 3 months and 5 months after birth. In addition, the growth of Meishan dolphin, subcutaneous fat, and the occurrence of BCL2 decreased gradually. The common phenomenon among varieties is that the growth rate of cells in subcutaneous fat is lower than that in immature cells, and the growth rate of cells in different varieties is lower than that in immature cells. In comparison with MSPA, the expression of prokaryotes in G1, S and M phases of MSPA was higher in Western species than in Chinese species.
