胃MALT型悪性リンパ腫におけるLセレクチンリガンド糖鎖の発現と機能解析

项目摘要

1.昨年度までの研究結果から,胃MALT型悪性リンパ腫の粘膜固有層に出現している高内皮細静脈様血管内腔面には,主にMECA-79抗体(-),HECA-452抗体(+)の糖鎖が発現していることが明らかになった。MECA-79(-)/HECA-452(+)の免疫組織化学的プロファイルから,この糖鎖は伸長型コア1構造上の非硫酸化シアリルルイスXであるごとが推測され,慢性活動性胃炎など,慢性炎症性疾唐で出現するMECA-79(+)/HECA-452(+)の通常のLセレクチンリガンド糖鎖どは異なるものである。この点は胃MALT型悪性リンパ腫の病理診断をする上で参考になりうる所見である。2.上記1.より,胃MALT型悪性リンパ腫では,シアリルルイスX糖鎖に硫酸基を付加する硫酸転移酵素の誘導が起こりにくい可能性が考えられた。そこで,パラフィン埋ブロックからRNAを抽出し,消化管粘膜での発現が報告されている硫酸転移酵素,GlcNAc6ST-1およびGlcNAc6ST-2のRT-PCRによる遺伝子発現解析を行った。GlcNAc6ST-1の発現は高率に検出された一方,GlcNAe6ST-2の発現はほとんど認められなかった。このことから,胃MALT型悪性リンパ腫の粘膜固有層ではGlcNAc6ST-1が発現しているが,効率的に伸長型コア1構造上のシアリルルイスXを硫酸化できない可能性が示唆された。現在までのところ,in situ hybridizationによる遺伝子発現の局在解析は成功していない。
