{目的}本研究は、動脈硬化性疾患のひとつであるヒト腹部大動脈瘤(AAA)組織検体およびAAAラットモデルにおいて、近年新たに線溶関連蛋白のレセプターとして同定されたアネキシンII(ANII)の発現状況を確認し、瘤進展の原因について考察しようとするものである。{研究方法}(1)同意の得られた人工血管置換術施行腹部大動脈瘤患者の術中切除標本について抗ヒトANIIモノクローナル抗体を用いて免疫組織学的検索を行った。染色強度は画像解析ソフトを用い、より客観的な半定量を行った。(2)Wister系雄性ラットを麻酔、開腹し腹部大動脈を露出、elastaseあるいは生理食塩水を還流し、後日大動脈組織を採取した。採取組織はそれぞれReal Time PCR法によるmRNAの定量および免疫組織学的検討に処した。{研究結果}(1)ヒト大動脈瘤検体では粥腫肩の部分にCD68陽性細胞に一致してANIIの発現亢進がみられた。繊維性プラークでは主にαSMA陽性細胞に一致してANIIの発現がみられた。(2)エラスターゼの還流時間を変えることにより、瘤径および弾性繊維の破壊の程度の異なる2種類のモデルを作成した。コントロールラットと比較して、瘤モデルラットでは中膜におけるANII、 tPAの発現が著しく亢進していた。また、ANIImRNAはコントロールと比較して、より瘤径の小さいモデルに有意に発現が亢進していたが、瘤径の大きなモデルでは有意差はみられなかった。一方、tPAは瘤径にかかわらずモデルラットに発現亢進がみられた。瘤径のより小さいラットがより初期の病変を反映していると仮定すれぱ、ANIIは瘤形成のより初期の段階に作用する可能性が示唆された。

In this study, patients with sclerosing diseases (AAA) were involved in major abdominal tumor (AAA). In recent years, we have studied the causes of major abdominal tumor (AAA). MRNA was used to quantify the immunological parameters. {results of the study} (1) after a lot of exercise, some of the CD68 sexual cells showed hyperactivity in ANII. The relationship between the main SMA and the sex cell is consistent with that of ANII. (2) to determine the duration of the flow, the degree of damage of the tumor and the size of the tumor. this is the result of a series of tests. There are signs of hyperactivity in the media, such as ANII and tPA. The size of the tumor is small, the diameter of the tumor is large, and the diameter of the tumor is larger than that of the ANIImRNA. On the other hand, the diameter of the tPA tumor showed hyperactivity. The diameter of the tumor was small, and the initial stage of the tumor was reflected in the early stage of the disease. ANII showed that the tumor was formed in the early stage, the possibility of tumor formation, the possibility of tumor formation, and the possibility of tumor formation.
