To clarify the possible roles of interactions with bone microenvironment in oncogenesis of odontogenic epithelium, expression of matrix-degrading proteinases, angiogenic factors, growth factors, and their associates was analyzed in ameloblastomas and malignancy ameloblastomas as well as in tooth germs.1. uPA, uPAR, PAI-1, and maspin: uPA was recognized predominantly in mesenchymal cells, uPAR was evident in epithelial cells, PAI-1 was found in both epithelial and mesenchymal cells, and maspin was expressed only in epithelial cells. Expression of uPA and uPAR tended to be higher in ameloblastic tumors than in tooth germs, while PAI-1 was slightly lower in ameloblastic tumors than in tooth germs. Maspin expression was significantly higher in ameloblastomas than in tooth germs.2. PD-ECGF/TP and angiopoietin1, 2: PD-ECGF/TP was expressed in mesenchymal cells, and showed higher reactivity in ameloblastomas than in tooth germs. Angiopoietin1 and 2 were expressed mainly in epithelial cells neighboring the basement membrane, and malignant ameloblastic tumors showed low angiopoietin1 expression and high angiopoietin2 expression as compared with tooth germs and ameloblastomas.3. IGF, PDGF, and their receptors: These molecules were expressed mainly in epithelial cells neighboring the basement membrane. Expression of IGF-II was higher in ameloblastomas than in tooth germs, and desmoplastic ameloblastomas showed high expression of IGF and the receptor. Malignant ameloblastic tumors showed high expression of PDGF and the receptor, and PDGF expression was higher in follicular ameloblastomas than in plexiform ameloblastomas.
