植物のRNAサイレンシング誘起の初期過程において、small interfering RNA (siRNA)は2本鎖RNA切断酵素DCLの働きにより作られ、HEN1によりメチル化される。DCLとHEN1が複合体を形成している可能性を検討したが、結果は否定的であった。これは、トマトモザイクウイルスの複製タンパク質が、生成後遊離した非メチル化2本鎖siRNAを競合的に奪取して、HEN1によるメチル化を介したRNAサイレンシング経路への流入を阻害している可能性と矛盾しない結果であった。また、試験管内合成したAGO1タンパク質が細胞抽出液中に存在する因子の助けを借りて遊離の2本鎖siRNAを取り込み、siRNA配列に特異的な標的RNA切断活性を現しうることを示した。
