平成19年度実績1.胃癌臨床検体の採取胃癌臨床検体を当院の胃内視鏡検査を受ける患者もしくは手術を受ける患者から収集中である。採取時には倫理委員会ですでに承認の得られている同意説明文書にて本研究の主旨を十分に説明し文書同意を得た後、検体はホルマリン固定もしくはTissueTechに包埋後、直ちに液体窒素で凍結し保存した。2.高密度オリゴヌクレオチドアレイを用いたゲノム解析1.のごとく採取した検体からマイクロダイセクション(ライカ社)を用いて癌部・非癌部を選択的に切り出し、回収したサンプルよりゲノムDNAを抽出した。今後は複数の臨床検体で共通の遺伝子増幅および欠失領域やLOHの領域を網羅的に解析し、さらにその結果を臨床・病理学的パラメータと組み合わせることにより、癌の進展・予後予測との関連性を検討する。3.癌遺伝子および癌抑制遺伝子候補の分子生物学的解析2.のごとく胃癌細胞株35種を用いて解析した結果、ゲノム異常の頻度が高いなど胃癌の癌遺伝子および癌抑制遺伝子の候補と考えられる遺伝子について、臨床検体を用いて解析を行なった。細胞株で増幅やLOHが高頻度に起きている領域は、臨床検体でもその変化が高頻度に認められ、細胞株の異常が臨床検体の異常を反映していることが示唆された。今後これらの領域に含まれる遺伝子の胃癌におけるタンパク発現やその機能など、胃癌の発生や進展にかかわる生物学的な意義について解析を行なう。
