間質性肺炎群と健常人の血清および気管支肺胞洗浄液を用いてTSP-1濃度をELISA法にて測定した。血清TSP-1濃度は間質性肺炎群で高値を呈し、VEGF濃度と相関しており、本疾患の病態におけるTSP-1の重要性が示された。免疫組織化学的検討では再生肺胞上皮細胞および肺胞マクロファージの重要性が示唆された。TSP-1に対するsiRNA(short interfering RNA)を数種類作製し、気道上皮細胞を用いた実験において、一つのsiRNAで良好な抑制効果がみられるようになった。肺線維症動物モデルにおける気道へのsiRNA-TSP-1投与方法を思案しながら、抑制実験にとりかかっている。
