Altered renal medullary gene expression to regulate pressure-natriuresis in diabetic hypertensive rats

研究了糖尿病高血压大鼠（OLETF大鼠）及其对照大鼠（LETO大鼠）中与氧化应激和成骨相关的基因表达。骨形态发生蛋白2（BMP 2）、核心结合因子1（cbfa 1）、基质gla蛋白（MGP）、碱性磷酸酶（Alp）、骨γ羧基谷氨酸蛋白2（Bglap 2）、过氧化氢酶、Gp 91 phox、通过实时逆转录分析肾髓质和皮质中的p22 phox和klotho。与LETO大鼠相比，OLETF大鼠肾髓质中p22 phox和Gp 91 phox mRNA的基因表达增加。与LETO大鼠相比，OLETF大鼠肾皮质BMP 2、cbfa 1、MGP和Alp mRNA表达增加。与LETO大鼠相比，OLETF大鼠肾髓质中klotho mRNA减少。OLETF和LETO大鼠肾脏髓质和皮质的过氧化氢酶mRNA表达无明显差异，这些基因表达的改变可能与糖尿病高血压大鼠压力-尿钠排泄的逆转和心血管钙化的增加有关。

Gene expressions related to oxidative stress and osteogenesis were investigated in diabetic hypertensive rats (OLETF rats) and its control rats (LETO rats). Gene expressions of bone morphogenetic protein 2 (BMP2), core binding factor 1 (cbfa1), matrix gla protein (MGP), alkaline phosphatase (Alp), bone gamma carboxyglutamate protein 2 (Bglap2), catalase, Gp91phox, p22phox and klotho in renal medulla and cortex were analyzed by real-time reverse transcription-polymerase chain reaction.Gene expressions of p22phox and Gp91phox mRNA were increased in the renal medulla of OLETF rats compared with LETO rats. BMP2, cbfa1, MGP and Alp mRNA expression were increased in the renal cortex of OLETF compared with LETO rats. klotho mRNA in the renal medulla of OLETF rats was reduced compared with LETO rats. Catalase mRNA expression of both renal medulla and cortex did not differ between OLETF and LETO rats.These altered gene expression may contribute to rightward shift of pressure-natriuresis and enhancing cardiovascular calcification in diabetic hypertensive rats.
