Gene analysis on the osteoblast proliferation and differentiation by culturing with biomaterial derived from natural resources.

用总浓度为0.1%(w/v)的鱼胶原蛋白(α-MEM)培养人成骨细胞。用细胞计数法检测成骨细胞的增殖情况，用实时荧光定量聚合酶链式反应和免疫印迹法检测矿化相关基因和蛋白的表达。此外，通过von Kossa染色观察FC对成骨细胞的作用。结果显示，与对照组相比，FC组细胞增殖和矿化相关基因和蛋白表达增加。延长孵育时间后，FC组矿化结节较对照组增多。

A human osteoblast was cultured with α-MEM containing fish collagen (FC) that was total concentration of 0.1%(w/v). Osteoblast proliferation by cell count and the expression of mineralization-related genes and proteins with real-time PCR and western blot analyses were investigated. Furthermore, von Kossa staining was performed to study the effect of FC on the osteoblast at the prolonged culture. The results revealed that cell proliferation and mineralization-related genes and proteins increased in the FC group compared with control group. After the incubation of prolonged period, mineralized nodule of FC group augmented than that of control group.
