We isolated circulating tumor cells(CTCs) using CellSearch^<TM> in five cases of gastric cancer patients with disseminated carcinomatosis of bone marrow. Then, we could identify several candidate genes involved in pathogenesis of this disease, based on the result of gene expression profiling of collected CTCs. As CTCs in two patients remarkably increased and most of CTCs highly expressed HER2, we used' trastuzumab', the monoclonal antibody against HER2, combined with the chemotherapy, against these two patients. They had remission of DIC and long-term survival. Moreover, we could establish mouse xenograft model using CTCs obtained from two gastric cancer patients with disseminated carcinomatosis of bone marrow.

我们使用细胞搜索^<tm>在五例胃癌患者中分离出循环肿瘤细胞（CTC）。然后，我们可以根据收集的CTC的基因表达分析的结果来鉴定几个参与该疾病发病机理的候选基因。随着两名患者的CTC明显增加，大多数CTC高度表达的HER2，我们使用了对HER2的单克隆抗体的“曲妥珠单抗”，并与化学疗法结合了这两名患者。他们缓解了DIC和长期生存。此外，我们可以使用从两名胃癌患者获得的CTC建立小鼠异种移植模型。
