Investigation of stretch-induced intracellular signal transduction in human periodontal ligament fibroblasts exposed to periodontopathic factors.

In this study, we investigated the mechanism of intracellular signaling cascade with mechanical stress using the human periodontal ligament fibroblast(HPLF). And we investigated the cellular response of the active center of lipopolysaccharide(LPS) endotoxin which is a virulence factor of periodontal disease. Moreover, we researched the effect of mechanical stress against these cell receptors. As a result, we confirmed that HPLFs were expressed on the inflammatory cytokines via the MAPK signaling cascade in the uni-axial stretch. Next, we found that the expression of inflammatory cytokines through the MAPK signaling cascade was enhanced by the mechanical stress after stimulating by the virulence factors of periodontal disease.Therefore, it is suggested that the cell receptor against the center of virulence of LPS might be affected by the mechanical deformation. Further study is required in the future.
本研究以人牙周膜成纤维细胞(HPLF)为研究对象,探讨机械应力对细胞内信号级联反应的影响机制。并探讨了牙周病毒力因子脂多糖(LPS)内毒素活性中心的细胞反应。此外,我们还研究了机械应力对这些细胞受体的影响。因此,我们证实了在单轴拉伸中,HPLFs通过MAPK信号级联在炎性细胞因子上表达。其次,我们发现牙周病毒力因子刺激后,机械应力可通过MAPK信号级联途径增强炎症细胞因子的表达,提示机械变形可能影响了LPS毒力中心的细胞受体。今后需要进一步研究。
