项目摘要

(1)Adiponectin投与にてAPC欠損(Min)マウスの小腸adencmaの個数と大きさは有意に低下した。ヒト、胃癌、大腸癌組織におけるadiponectin受容体(AdipoRI and R2)のmRNA,タンパクの発現量をRT-PCR免疫染色で検討したところ、健常部と比べて共に有意に低下していた。(2)胃培養癌細胞において、TGF-β添加がAdipoR1とAdipoR2は有意に低下した。(3)Adiponectinは通常培養液で大腸癌細胞の増殖を抑制したが、グルコース欠乏状態では、細胞の生存を補助する役割を果たし、これはautophagyを介していることが判明した。以上より、腹腔内脂肪由来のadiponectinは癌の発生、進展を制御する環境因子として重要な役割を有している。一方、TGF-βはadiponectinの受容体を低下させることによって、その抑制作用を回避させる機序を介して癌の進展に促進的な作用を発揮することが推測された。(4)胃癌術後の内臓脂肪量と皮下脂肪量の変化を臍レベルのCT画像を用いたFATScanにてretrospeotiveに検討すると、迷走神経温存群と切離群との間で、皮下脂肪量の変化には差を認めなかったが、内臓脂肪量は迷走神経温存群で優位に高値に保たれていた。迷走神経は腹腔内脂肪を調節する生理学的機能を有している事が示唆された。

(1)Adiponectin administration and APC deficiency (Min) The number of adencma in the small intestine is large and intentionally low. Adiponectin receptors (AdipoRI and R2) mRNA expression was detected by RT-PCR immunostaining in human gastric, colorectal and gastric cancer tissues. (2)Cultured gastric cancer cells were intentionally down-regulated by addition of AdipoR1 and AdipoR2 with TGF-β. (3)Adiponectin normal culture medium can inhibit the proliferation of colorectal cancer cells. These findings suggest that adiponectin is an important environmental factor in the development and progression of cancer. One of the mechanisms involved in the inhibition of TGF-β adiponectin receptor downregulation and its role in promoting cancer progression was postulated. (4)The change of internal fat mass and subcutaneous fat mass after gastric cancer operation is discussed in the FATScan, retropetive examination, and the change of subcutaneous fat mass is recognized in the FATScan, and the change of internal fat mass is maintained in the optimal position of the FATScan. The physiological function of the brain in regulating fat in the abdominal cavity has been demonstrated.
