Das gemeinsame Thema dieser Zusammenarbeit ist die Mobilität von makromolekularen Komplexen in lebenden Zellen und ihre Interaktion mit aktivem oder inaktivem Chromatin. Unsere vorläufigen Daten zeigen, daß einige GFP Fusionen von chromatindichten Zellkernregionen ausgeschlossen sind und daß diese ungleiche Verteilung aktiv erhalten wird. Wir wollen jetzt untersuchen, ob diese Verteilung von ihrer Größe, Ladung oder Affinität abhängt. Ferner soll untersucht werden, ob diese räumliche Proteinverteilung mit unterschiedlichen Diffusionsraten korreliert und welche Eigenschaften der Chromatinorganisation dafür verantwortlich sind. Wir wollen den Effekt von Transkriptions-aktivierung, DNA Methylierung und Histonmodifikation auf Chromatinstruktur und Zugänglichkeit untersuchen. Wir werden induzierbare Transgene mit zwei verschiedenen Operatorarrays zur Markierung mit grün und rot fluoreszierenden Proteinen herstellen. Die Verwendung von zwei Fluorophoren ermöglicht eine präzise Distanzmessung jenseits der optischen Auflösungsgrenze. Ferner sollen ganze Chromatindomänen markiert werden um deren Größe und Zugänglichkeit in Abhängigkeit von DNA und Histonmodifikationen zu untersuchen. Verschiedene Modifikations-enzyme sollen spezifisch zu diesen markierten Domänen rekrutiert werden. Diese Kooperation soll zum Verständnis der Prinzipien und Effekte der funktionellen Genomorganisation beitragen. Summary: The common goal of this collaboration is to determine the mobility of macromolecular complexes in living cells as well as their access to and their interaction with transcriptionally active or inactive chromatin. Our preliminary data indicate that some GFP fusion proteins are excluded from chromatin dense regions of the nucleus and that this uneven distribution is actively maintained. Now, we want to study whether access to chromatin depends on size, charge or affinity. In addition, we will investigate whether the uneven distribution of proteins in the nucleus correlates with spatial differences in diffusion rates. In parallel, we want to investigate which features of chromatin organisation are responsible for this exclusion. We want to assess the effect of transcriptional activation, DNA methylation and histone modifications on chromatin organisation and accessibility. We will generate inducible transgenes featuring two different operator arrays labelled with green and red fluorescent binding proteins. The use of spectrally distinct fluorophores will allow a precise distance measurement beyond the optical resolution limit. Larger arrays will be used to labelled entire chromatin domains and to determine their size and accessibility as a function of DNA and histone modifications. Different modifying enzymes will be specifically recruited to these labelled domains via the operator arrays. This collaborative project should help to elucidate the principles and effects of functional genome organisation.
