项目摘要

自己複製能と多分化能をもつ癌性幹細胞は、腫瘍細胞集団中にごく少数存在し、癌の再発・転移や治療抵抗性に大きく関与していると考えられている。その本態の解明と治療抵抗性の克服は、治療成果の飛躍的な改善につながると期待されている。しかし、非常に難治性で予後不良な成人T細胞白血病/リンパ腫(ATLL)において、未だ癌性幹細胞に関する報告は無い。そこで、癌性幹細胞の観点からATLLの多段階発癌機構を解明し、ATLL発症高度危険群を同定する検査法と有効な治療法の開発を目的として、研究を進めている。癌性幹細胞が、原因ウイルスHTLV-1感染やATLLの発症・進展にどのように関与しているのか、造血細胞特異的で腫瘍化に関与するSHP1と活性酸素種、幹細胞特異的マーカー、T細胞フェノタイプマーカーを用いて、検出を試みた。まず、癌性幹細胞が腫瘍細胞中に存在するのかどうか、コントロールとして正常リンパ節、ATLL細胞株数種(患者検体由来、IL2依存性/非依存性)を用いて、免疫染色を行った。通常SHP1は細胞膜上に存在しており、増殖等を抑制する機能を持つため、正常リンパ節では、休止期を示唆する成熟リンパ球が陽性を示していた。これに対して、腫瘍細胞ではSHP1発現が低下し増殖が亢進していることを、我々はこれまでに報告している。しかしながら、ATLL細胞株においてSHP1陽性所見を認め、さらに正常リンパ球とは局在が異なっていることがわかった。このSHP1の局在の差異と活性化状態の関連や、ATLLに特徴的なFlowercellとの関連についても検討している。また、癌性幹細胞の指標として特異性が高いとされるアルデヒド脱水素酵素(ALDH)を追加した。ALDHの数種のアイソタイプを認識する抗体を用いて2重染色を行い、癌性幹細胞のプロファイルの解析を進めている。

SHP 1 is usually present on the cell membrane, inhibiting growth, etc., and maintaining its function, normal release, resting period, mature release, and positive indication. In addition, SHP 1 expression in tumor cells was decreased and proliferation was increased. ATLL cell line SHP-1 positive was detected, and normal cells were detected. The difference between SHP 1 and its activation state is related to ATLL, and the correlation between SHP 1 and ATLL is related to ATLL. In addition, the specificity of cancer stem cell markers was increased. Several types of ALDH are recognized by antibodies and analyzed by two-fold staining.
