皮膚細胞混濁液に含まれる細胞数を同量にしても皮膚再構築は起こらなかったことより、まず細胞腫に違いがある可能性を考えた。週齢により上皮の厚さや真皮における細胞密度に違いはあり、フローサイトメトリー（CantoII）を使用し細胞腫の割合を比較検討した。胎生18日と4週齢のGFP導入C57Bl/6Jマウスの皮膚をhomogeniseした後、0.1% collagenase1(2%BSA in DMEM)で37℃1時間反応させ皮膚細胞混濁液を作成した。それぞれChamber法でC57Bl/6Jマウス（オス、8週齢）の背部皮膚潰瘍部にそれぞれ移植する際。フローサイトメトリーの結果、細胞数に明らかに胎生１８日の皮膚細胞混濁液に多く含まれた違細胞を４週齢の皮膚細胞混濁液に追加し移植実験を行った。
