本研究の目的は、ハタ科魚類の種苗生産を安定的に行うための一助として、卵黄形成期の雌に特異的に出現する卵黄前駆体タンパク(ビテロジェニン)を指標とした成熟判別ならびに雌雄判別法を確立することである。しかし、本科魚種のビテロジェニンは精製されておらず、その検出系も確立されていない。そこで、まず雌性ホルモンを投与し人為的にビテロジェニンを誘導したキジハタの血液より、ハイドロキシルアパアタイトクロマトグラフィーおよびゲル濾過クロマトグラフィーを用いてビテロジェニンを分離精製した。その結果、純度の高いビテロジェニンの精製に成功した。これを家兎に免疫してビテロジェニン特異抗体を作製した。この抗体をキジハタ雄血清と混和し、雌雄共通のタンパクに対する抗体を吸収除去した。この吸収抗体はビテロジェニンとのみ反応することが確認された。次に、本抗体を用いたキジハタ卵黄前駆体タンパクの検出系の開発を行った。検出系には増養殖の現場での実用性を考慮に入れ、多量の検体を短時間で容易に処理できるドットブロッテイング法を選んだ。その結果、血中ビテロジェニンは19.5ng/dotまで検出できることが分かった。これは、本検出系が従来のマンシーニ法による検出系より高感度であることを示唆している。また、雄血清とは全く反応しなかったことから、雌雄判別に有効な方法であることが明らかとなった。最後に、本検出系が他のハタ科魚類のビテロジェニン検出にも用いることが出来るか否かを調べるため、マハタ・クエとの免疫交差性を調べた。その結果、何れのビテロジェニンとも交差する事が分かった。本研究により、ハタ科魚類の雌雄判別をビテロジェニンを指標として行うことが可能になった。本研究の成果は、平成9年度日本水産学会春季大会(東京)にて発表する予定である。また、近日中に、論文としても公表する予定である。
