We succeeded in differentiating different donor-iPS-derived hepatoblasts by using the same protocol. Their hepatoblast characters were confirmed by both protein (immunostaining, flow cytometry) and genetic expression (qPCR analysis).Nonetheless, there is some variance in the expression of hepatoblast markers, which implies intra-strain differences between these strains. Each hepatoblast albumin secretion is also confirmed by protein assay and the maturation into hepatocytes is verified by hepatocyte differentiation. All cells possess the ability to proliferate in vitro and largely expanded throughout the passages. These cells are then transplanted into the chronic hepatic injury model.
