We investigated Gallyas-positive glial structures immunohistochemically in cerebral cortex of patients with diffuse neurofibrillaly tangles with calcification (DNTC). DNTC is characterized by slowly progressive cortical dementia in the proscenium, localized temporal or temporofrontal lobar atrophy, numerous neurofibrillary tangles widespread in the cerebral cortex, and pronounced calcareous deposits. In addition to neurofibrillary tangles, a variety tau-positive glial structures occur. Double immunostaining demonstrated that thorn-shaped astrocyte are located in cerebral white matter with severe gliosis and that agglomerate and amorphous argyrophilic structures of partially tau-positive fibers are found in fronto-temporo-parietal cotices and subcortical white matter. Glial coiled body-like structures in cerebral white matter are not stained by teansferrin. Totally tau-positive argyrophilic structures are found numerously in front-temporo parietal cerebral cortex and white matter. Partially tau-positive argyrophilic structures stained by GFAP are recognized in fronto-temporo-parietal cotices. Argyrophilic end-feet are found in regions with severe gliosis. These results suggest that formation and destribution of Gallyas/tau-positive glia in this disorder are different form other types of degenerative disorder with dementia.
