Local Chemotherapy via Hepatic Arterial and Portal Blood Flows For Liver Cancer

(a)经兔肝动脉接种VX2肿瘤，同时经门静脉注入1mg/kg剂量的ADM （Gl（+）组（n=6））。对照组（Gl（-）组（n=6））只注射ADM，不注射谷胱甘肽。然后认真分析两组大鼠肝组织中ADM浓度。分别于给药前、给药后5、10、15、20、25、30、45分钟取肝组织及腔静脉血，采用高效液相色谱法测定ADM浓度。(b)在同一模型下，比较两组的抗癌效果。(c)（结果）(a) (1) Gl（+）组和Gl（-）组ADM浓度相同，均在ADM给药后5 ~ 10分钟达到最高值。(2) Gl（+）组肝脏中ADM浓度高于Gl（-）组。(3) Gl（+）组与Gl（-）组血中ADM浓度无显著性差异。(b) Gl（+）组动物的中位生存日为18.6±7.3天，Gl（-）组动物的中位生存日为16.4±6.3天。

(a) Via the hepatic artery of the rabbits inoculated VX2 tumor, ADM at a dose of 1mg/kg was administered and GSH was also infused via the portal vein at the same time (Gl(+)group (n=6)). As a control group, the rabbits were administered only ADM without GSH infusion (Gl(-)group (n=6)). Then the ADM concentrations in the liver tissue in both groups were analyzed seriously. Before and 5, 10, 15, 20, 25, 30 and 45 minutes after the ADM administration, hepatic tissue and the blood in the caval vein were sampled and measured the ADM concentration by the HPLC method.(b) In the same models, anticancer effect in both groups were compared.(c) (Results) (a) (1) The ADM concentrations showed the same values, with the highest in 5 to 10 minutes after ADM administration in both Gl(+) and Gl(-) groups.(2) The ADM concentrations in the liver of Gl(+) were higher than those of Gl(-) group.(3) There was no difference between the ADM concentrations in the blood in Gl(+) and Gl(-) groups.(b) Median survival day of the animals in the Gl(+) group was 18.6±7.3, and that of Gl(-) was 16.4±6.3 days.
