The central nervous system anomalies induced by anti-pregnant rat kidney serum (ARKS). In order to clarify the mechanism of immune induced anomalies in rat, we studied several numbers of categories. The results of study related to oncogene, we could not demonstrate an expression of s-myc, c-, nyc, fas and RB.Only wild type of P53 was expressed, and it was considered that this supressor gene may be induced to repair the damaged nuclei by ARKS.On the tumor growth factor, Mib-l and Ki-67 did not express and only PCNA revealed higher in control which were injected saline instead of antiserum than ARKS injected rats. This results suggested that the ARKS inhibit the growth of epithelial cells of the neural tube. The abnomality of chromosome was not detected at this time. The cell mediated immunity did not act directry but act indirectry to the fetus which treated by ARKS.Fluorescence immunohistochemical studies were performed to localize the binding of two lectins, concanavalin A (Con A) and wheat germ agglutinin (WGA), to glycoconjugates on neuroepithelium. We found fluorescence was observed on the luminal side of the neuroepithelium in the intercell- ular region and on the basement membrane of the neural tube in embryos on day 9 of gestation (GD 9). In GD 21 embryos there was much more intense fluorescence in the extracellular matrix and the ependymal lining cells of the cell surface of the neuroepithelium during neurulation was different in rat embryos injected with ARKS than in normal embryos injected with NRS.These results support the idea that simple nonclosure and overgrowth constitute the mechanism of neural tube defect.
