われわれは5リポキシゲナーゼ遺伝子プロモーター領域に興味深い遺伝子多型を発見し、それが遺伝子の発現調節に関わっていることを明らかにした。本研究の目的は、日本人健常者および喘息患者についてこの遺伝子多型と喘息の病型、重症度、治療に対する反応性との相関を検討することにより、同遺伝子多型の臨床的意義を明らかにすることである。慶應義塾大学病院ならびに関連病院に通院中の喘息患者211例、健常人93例についてSSLP(Simple sequence length polymorphisms)法による5リポキシゲナーゼ遺伝子プロモーター領域遺伝子多型解析を行った。その結果、日本人においても同多型が存在すること、しかしその発現頻度は白人種、黒人種のいずれとも明らかに異なることが明らかとなった。さらに病型、重症度など臨床データの解析により、特定のアレルの発現頻度が小児喘息の既往がありダニ抗原に対する特異的IgEが陽性である症例において高いことを見いだした。Irvinらは5リポキシゲナーゼ欠損マウスにおいて抗原特異的IgG、IgE産生が抑制されることを報告しており (Am.J.Physiol.1997)、我々の結果との関連が興味深い。今後さらにこの遺伝子多型とロイコトリエン受容体拮抗薬に対する反応性との関連について解析を行っていく予定である。

In this paper, we discuss the development of genetic polymorphism in different regions, and discuss the regulation of genetic polymorphism. The purpose of this study is to investigate the correlation between this genetic polymorphism of wheezing in Japanese healthy adults and wheezing patients and the disease type, severity, and treatment, and to clarify the clinical significance of the genetic polymorphism. The SSLP(Simple sequence length polymorphisms) method was used to analyze the genetic diversity of 211 wheezing patients and 93 healthy subjects in Keio University Hospital and related hospitals. As a result, Japanese people have the same polytype, and the frequency of occurrence of white people and black people is different. The disease type, severity, clinical characteristics, frequency of occurrence of specific diseases, history of wheezing, antigen-specific IgE, and positive symptoms are all high in severity. Irvin has been reported to be interested in inhibiting the production of antigen-specific IgG and IgE. Am.J.Physiol.1997 In the future, this gene polymorphism will be analyzed in the context of receptor antagonism.
