This core facility will provide support with regard to flow cytometric analysis and flow cytometric cell separation for a number of projects within the proposed center. Three machines are currently accessible: two bench-top analysis machines and one high-speed 8-color, 10-parameter Moflo sorter with single-cell deposition unit and four-way sorting capabilities. An additional sorter (FACSAria) might be purchased in the future depending on availability of funding. To increase access to this limited resource, a second technician was recently hired to increase the number of hours the machines are accessible for use. One-third salary support is requested for this technician. The facility will provide a small panel of titrated reagents for typical flow cytometric analysis of murine, human and baboon cells. Training, data archiving and data evaluation will also be provided. Projects 1 (Aims 3 and 4), Project 2 (Aims 1-3) as well as Technology Development Component projects 2 and 3 require access to flow facilities. This includes particularly the sorting of GFP-tagged myeloid cells after retroviral infection as well as sorting of starting populations, such as human alveolar macrophages and peripheral blood macrophages. FACS-analysis is required in particular for apoptosis detection of macrophages and dendritic cells, for PMN functional tests and for the analysis of lymph node, spleen and peripheral blood samples from B. anthracis-infected baboons.
