This project will aim to build a core fundamental understanding of the theory relevant to this membrane-like support structure and its application in biological target capture via adsorption and elution via desorption. The aim being to guide input into the second generation of the Fibro technology platform. With this fundamental knowledge in place, known production methods will be used to generate samples at Cytiva facilities to explore and verify this understanding. The project will rely heavily on multi factorial Design of Experiment approaches to identify the key parameters that impact subsequent performance characteristics like binding capacity, back pressure and fouling. A goal of the project will be to include this into known mechanistic models which supports the use of the technology and provides fundamental understanding and prediction of improved designs. While much of the initial verification work will utilise model proteins and mAbs, the knowledge built in this project is also intended to be relevant for larger biologic targets like viral vectors.
