CDT Theme alignment: Advanced Product Design Transport processes for synthetic and natural macromolecules are not fully understood, but govern whether a drug is delivered in the right place, at the right time, and the right dose. This project will look at new synthetic and natural macromolecules and probes for real-time reporting of delivery. We will use FRET and related optical proximity probes between fluorescently labelled macromolecules and selected 'landmarks' of their environment, to gain information on localization in biological settings (tissue and cell) at the molecular level. The project will encompass synthesis of new responsive and biodegradable polymer delivery systems, conjugation of fluorescent probes, evaluation of delivery system and cargo transport in cell / tissue culture, and potentially in vivo experiments. Finally, we aim to exemplify the new materials and knowledge of trafficking to enhance therapeutic efficacy of siRNA knockdown in acute kidney injury (AKI)
