The ability to detect, respond to and effectively repair tissue damage following injury or infection is critical to an organism's long-term survival. Integral to this is the interaction between inflammation and pain, through the sensitisation of nociceptive pathways in the injured or infected tissue. Behaviourally, this protective response manifests as hyperalgesia and allodynia through the activation and sensitisation of pain-sensing neurones to thermal, chemical and mechanical stimuli. Multiple mediators are released by resident or recruited blood-derived immune cells that are capable of sensitising pain pathways, and of these, cytokines play a critical role. However, in many cases, the mechanisms by which cytokines induce pain and sensitise nociceptors are poorly understood.
