Magic-Angle-Spinning Dynamic-Nuclear-Polarization (MAS-DNP) solid-state NMR is beginning to be used to study complex biological structures and biomaterials. Kockenberger/Thomas propose to undertake pioneering studies that are intractable using other methods:A. Using the polarization agent AMUPol, the structure of the crystalline and amorphous regions of 4RepCT spider silk will be studied to identify key aa interactions and correlate these with mechanical properties.B. 4RepCT incorporating L-azidohomoalanine will be conjugated with Gd(III)DOTA/nitroxide as polarization sources to study fibre self-assembly.C. A new targeted polarization strategy will be developed, based on the cell-wall binding domain of lysostaphin-Gd(III)DOTA to probe S. aureus cell envelopes.
