We aim to investigate how genetic variation impacts PSG gene regulation and consequently on cellular phenotypes in trophoblast and senescent cells. We will leverage cutting-edge stem cell and sequencing technologies with epigenomics and functional assays, bringing together expertise in trophoblast gene regulation (Branco lab), population genomics/epigenomics (Bell lab) and ageing (Altos Labs). More specifically, we aim to:1. Deeply characterise the genetics of the PSG locus on a set of induced pluripotent stem cell (iPSC) lines.2. Test the impact of PSG genetic variation on iPSC-derived trophoblast cells.3. Test the impact of PSG genetic variation on iPSC-derived cellular models of senescence
