"Heating-up" the immune microenvironment in paediatric high-grade glioma with combination immunotherapy

Immune cells have the potential to selectively eradicate high-risk paediatric brain tumours such as paediatric-type diffuse high-grade glioma (PDHGG). We aim to characterize the tumour immune microenvironment (TIME) of molecular subgroups of PDHGG, exploring immunological heterogeneity between tumours in different locations and with distinct molecular signatures. We aim to determine how this may be exploited to enhance efficacy of oncolytic virotherapy, a promising immunotherapeutic strategy capable of both direct tumour-cell lysis and immune stimulation. We aim to select immunotherapy combinations of oncolytic virus and molecular targeted therapeutics that can "heat-up" a "cold" TIME, identifying novel strategies for early phase trials.
