This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
Beta-two adrenergic agonists act to increase muscle mass in the elderly but the mechanism(s) in humans for this increase in muscle mass in unknown. We will examine the effects of 8.5 days on albuterol administration (16 mg/24 hours) on mixed muscle protein synthesis and on markers of muscle protein degradation (mRNAs for calpain-1 and calpain-2 and calpastatin). Additionally, we will measure IGF-I and IGF-II mRNA and the phosphorylation state of P70S6Kinase in elderly men (aged 60-85) who have body mass indices that correspond to the presence of sarcopenia. Elucidating the way in which beta two agonists affect muscle protein metabolism may possibly lead to other therapeutic strategies to target specific components of protein synthetic and/or protein degradative pathways that are activated by or deactivated by beta two agonists. Additionally, if we determine the primary site of action of beta two agonists we will be able to hypothesize whether beta two agonists likely work in aging related muscle wasting conditions.
