This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Two-hybrid screening is a powerful method to explore protein-protein interaction networks. For some organisms such as Plasmodium, it is difficult to make and screen two-hybrid libraries due to extremely high AT-content of Plasmodium genome. We will use yeast two-hybrid libraries to screen for interactions with several P.falciparum genes which have yeast homologs implicated in an array of different cellular processes. Plasmodium proteins homologous to the identified yeast partners will then be cloned and tested for interaction with plasmodium baits in two-hybrid system and by biochemical methods.
