This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This GCRC-supported project entails addressing Specific Aim 4 of the NIH grant: Specific Aim 4: To evaluate the potential of CBV, CBF and vascular morphology measurements to track tumor growth, progression and response to treatment in brain tumor patients. We demonstrated the feasibility of using a novel GE/SE imaging method to obtain measures of total (GE) and microvascular (SE) CBV, along with vessel size information (DR2*/DR2). The GE CBV and ratio showed significant differences between low and high-grade tumors, a result dependent on the proper consideration of contrast agent extravasation effects. Now, we propose to obtain multiparameter data sets (CBV, CBF, vessel caliber) from brain tumor patients over time and before and after radiation therapy, and compare them to relevant immunohistochemical (IHC) markers. We hypothesize that each parameter will play a unique yet complementary role in predicting tumor angiogenesis, tumor progression and response to therapy.
