This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
The recovery of broadband (650nm~1000nm) diffuse optical spectra stratified by layer in a tissue-simulating, two-layer phantom is reported for the first time. The broadband optical properties of the phantom featured a top layer consistent with lipids and a bottom layer consistent with muscle. The concentrations of chromophores in both layers (top layer thickness greater than 5mm) were calculated from the recovered diffuse optical spectra with an average 15% error. The recovery is based on a Fourier transform solution to the diffusion equation using multi-distance broadband diffuse optical reflectance measurements. The algorithm recovers the absorption and reduced scattering coefficients in both layers and top layer thickness. This algorithm is insensitive to initial guesses of both layers' optical properties as long as the layer thickness initial guess is within ¿ 2mm. The robustness of the fitting routine is further tested with Monte Carlo simulations.
