This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We have reconstructed 2-dimensional images of gene expression for 20,000 genes in a coronal slice of the mouse brain. The coronal section was at the level of the striatum and was analyzed using microarrays and voxelation, in which the brain is divided into spatially registered cubes or voxels. The resolution was 1 mm3. Known and novel genes were identified with expression patterns localized to defined substructures within the brain. In addition, genes with unexpected patterns were identified and cluster analysis identified a set of genes with a gradient of dorsal/ventral expression not restricted to known anatomical boundaries. Voxelation analysis of the same section using LC-FTICR revealed good agreement with the transcript data. Here, we propose to create 3D voxelation maps of protein expression for the entire normal mouse brain and brains from mouse models of Parkinson's disease (PD). The resulting maps will be a valuable tool for the neuroscientific community.
