This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
Every electron micrograph is modulated by the microscope contrast transfer function (CTF) and associated microscope parameters during the image formation process. Since the CTF modulations are different for different micrographs, it is essential to accurately estimate the CTF model parameters for each micrograph so that the particle images from different micrographs can be corrected for the CTF modulation and merged to achieve highest resolution 3-D reconstruction. We have used an optimization algorithm to allow the determination of these parameters reliably and automatically.
