Project Summary/Abstract
The PI holds a current NIGMS R35 research award 1R35GM147209 (Spatial genetics
investigation of multinucleated cells). Under this award, the PI is requesting supplemental
equipment funds that will be used to purchase MERSCOPE, the first and only commercial
platform for Multiplexed Error-Robust Fluorescence in situ Hybridization (MERFISH) technology,
which allows the simultaneous single-molecule imaging of hundreds of mRNA species in
hundreds of thousands of cells. The MERSCOPE Platform also offers a suite of software tools
and optimized reagents that streamline the whole experiment process from sample to high-quality
spatial transcriptome data. It thus reduces human interventions and procedural errors. The
MERSCOPE system will serve as the workhorse for the parent R35 project. The main goal of the
funded R35 grant is to perform in-depth spatial investigations of the basic genetic mechanism
operated in gigantic syncytial cells like myofibers. These unique cellular systems contain
hundreds of nuclei and currently little is known about the interplay and coordination of gene
expression programs in heavily multinucleated cells. This administrative supplement, if granted,
will facilitate the purchase of a MERSCOPE Platform, which will serve a critical role in the success
of the funded R35 project. The Institutional supports were acquired with funds ready to purchase
service contracts throughout the life of the parent R35 project.
