This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. SPECIFIC AIMS 1. To test the effectiveness of broccoli sprouts in preventing oxidative-stress mediated respiratory inflammation through the upregulation of Phase II antioxidant enzymes. We will assess biomarkers of oxidative stress in the upper and lower airway as measured by upper and lower airway cytokine and 8-isoprostane levels and by GSTM1 and NQO1 gene expression. 2. To examine the role of GSTM1 and NQO1 genotypes, common polymorphisms we have shown to be critical in the adjuvant response to diesel exhaust particles (DEP), in determining responsiveness to broccoli sprouts. 3. To investigate the effects of upper airway oxidative stress on inflammatory markers of the lower airway in subjects with atopy and mild intermittent asthma, thereby testing the interrelationship between upper and lower airway inflammation in response to DEP and response to broccoli sprouts. 4. To test the effectiveness of broccoli sprouts in preventing primary allergic sensitization to a respiratory neaoantigen administered with DEP as an adjuvant.
