This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. We would like to collect fluorescence lifetime data on a protein (blood coagulation factor XIa, FXIa) labeled with Alexa Fluor 594 as a donor and Alexa Fluor 647 as an acceptor (R0 = 85A). We hope to confirm previous steady-state FRET work that indicated the activation of the dimeric factor XIa (yielding FXIa) causes a major conformational change and resulting in a major decrease in the distance between the two active sites.
