This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
We would like to collect fluorescence lifetime data on a protein (blood coagulation factor XIa, FXIa) labeled with Alexa Fluor 594 as a donor and Alexa Fluor 647 as an acceptor (R0 = 85A). We hope to confirm previous steady-state FRET work that indicated the activation of the dimeric factor XIa (yielding FXIa) causes a major conformational change and resulting in a major decrease in the distance between the two active sites.
