项目摘要

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The purpose of this study is to obtain biologic materials from the blood and lungs of normal individuals to establish a set of normal range for various parameters. These will provide important information when applied to individuals with various pulmonary diseases, and will help in understanding of the etiology and pathogenesis of various lung diseases. Samples obtained from fiberoptic bronchoscopy in normal individuals will be used as follows: (a) macrophages obtained from bronchoalveolar lavage (BAL) will be used to evaluate efficiency of gene transfer using different adenovirus vectors; (b) studies will be conducted to evaluate strategies to decrease infection of macrophages by different strains of HIV; (c) macrophages will be evaluated in terms of pattern of cytokine release following infection with adenovirus vectors; (d) similar experiments as the ones described in (a) and (c) will be conducted with airway epithelial cells collected by brushing the lung airways; (e) lung epithelial lining fluid collected by BAL will be evaluated for anti-adenovirus antibodies, for various cytokines and for vascular endothelial growth factor levels; (f) collected airway cells will be evaluated for the various gene expression studies using microarrays for the effects of smoking in the expression of antioxidant and other genes in the cells lining the airways of the lungs (airway epithelial cells) and how these changes correlate with the frequency in mutation changes of genes that are relevant to lung cancer.
