This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This project will examine the hypothesis that a protein complex is assembled at the myocardial intercalated disk. The proposed protein complex is composed of the tyrosine kinases, ErbB2 and Src; protein phosphatases 1/ 2 and the p21 activated kinases, PAK1 and PAK2. This complex regulates the phosphorylation of connexin 43, a protein that mediates the ionic and electrical communication between adjacent cardiomyocytes. This hypothesis will be examined by confocal microscopy and co-immunoprecipitation, to assess protein- protein interactions within this complex. This study will utilize a rabbit of myocardial ischemia and a hamster model of dilated cardiomyopathy to examine this hypothesis.
