The regenerative capacity of skeletal muscle is due to a pool of stem cells, termed satellite cells, which are resident within adult skeletal muscle. Despite their integral role in muscle growth and regeneration, our understanding of the basic mechanisms regulating these cells has yet to be fully elucidated. In this proposal we will address some issues related to how muscle satellite cells are regulated in response to exercise and toxin-induced injury. We will begin by defining the role of a novel protein, Xin, within the muscle satellite cells and clarify its relationship with other known signaling proteins, such as Beta-catenin. Secondly, we will determine the factors which play a role regulating Xin within the muscle satellite cell population. Finally, we will evaluate the function of Xin within the whole animal in response to exercise or toxin-induced muscle injury. The results of these studies will enhance our understanding of the fundamental processes necessary for skeletal muscle hypertrophy and regeneration.
